Tofamark 5 Tablets TOFAMARK contains Tofacitinib, a selective oral inhibitor of the Janus kinase (JAK) family of enzymes, predominantly targeting JAK1 and JAK3, with functional selectivity over JAK2. By inhibiting intracellular JAK pathways, tofacitinib blocks STAT protein phosphorylation and dimerization, thereby halting signal transduction for multiple key pro-inflammatory cytokines, including interleukins (IL-2, IL-4, IL-7, IL-9, IL-15, IL-21) and type I and II interferons. This modulation suppresses lymphocyte activation, proliferation, and downstream inflammatory cascades responsible for autoimmune pathology. TOFAMARK is indicated for the treatment of moderately to severely active rheumatoid arthritis, active psoriatic arthritis, ankylosing spondylitis, and moderately to severely active ulcerative colitis.
