Trioday Tablets Trioday is a fixed-dose single-tablet antiretroviral regimen combining Tenofovir Disoproxil Fumarate (an NtRTI), Lamivudine (an NRTI), and Efavirenz (an NNRTI). Tenofovir disoproxil fumarate and lamivudine are intracellularly converted to their active phosphorylated forms, which compete with natural nucleoside substrates and terminate DNA chain elongation. Efavirenz non-competitively binds directly to HIV-1 reverse transcriptase, causing a conformational change that inhibits RNA-dependent DNA polymerase activity. Together, these active components inhibit viral transcription and stop HIV-1 replication. It is indicated as a complete once-daily regimen for the treatment of HIV-1 infection in adults and pediatric patients.
