TIBSOVO 250 Tablets TIBSOVO contains Ivosidenib, a small-molecule inhibitor targeting mutated isocitrate dehydrogenase-1 (IDH1). Mutated IDH1 catalyzes the abnormal reduction of alpha-ketoglutarate (α-KG) to the oncometabolite 2-hydroxyglutarate (2-HG), which blocks cellular differentiation and promotes oncogenesis. By selectively inhibiting mutated IDH1, ivosidenib decreases 2-HG levels, restoring cellular differentiation in IDH1-mutated cancer cells. It is indicated for the treatment of adult patients with susceptible IDH1-mutated acute myeloid leukemia (AML), cholangiocarcinoma, and grade 2 IDH-mutant glioma.
